The aim of this study was to screen methanol extract of Sorghum vulgare L. leaves to display potent antioxidant activity in vitro and total flavonoid contents in order to find possible sources for future novel antioxidants in food and pharmaceutical formulations. DPPH assay and reducing power assay was performed for confirm the antioxidant activity of the methanol extract of Sorghum vulgare L. leaves. The total flavonoid contents of methanol extract of leaves of Sorghum vulgare L. was also determined. Methanol extract of leaves of Sorghum vulgare L. was also subjected to preliminary phytochemical screening test for various constituents. The total flavonoid contents of methanol extract of leaves of Sorghum vulgare L. (1.44mg CE/gm extract) was found. Phytochemical screening of methanol extract of leaves of Sorghum vulgare L. revealed the presence of alkaloids, flavonoids, glycosides, tannins, saponins, terpeniods and absence of steroids. Methanol extract of leaves of Sorghum vulgare L. showed significant antioxidant activity in DPPH assay and reducing power assay. These findings also provide a rationale for further studies on isolation of active principles and its pharmacological evaluation
